高效液相*谱-荧光检测法测定人血浆中的匹伐他汀

PitavastatinbelongstotheclassofcoenzymeAreductaseinhibitors.Veryfewmethodsofassayingpitavastatinfromhumanplasmaareavailableinliterature.Ananalyticalmethodispresentedforthedeterminationofthedrugfromhumanplasmamakinguseofthefluorescentpropertyofthedrug.Thedrugisextractedfromplasmausingethylacetateunderneutralconditionandthenanalyzedbyreversed-phasehighperformanceliquidchromatography(HPLC)withfluorescencedetection(λEx245nm;λEm420nm).AnalysisofpitavastatinwascarriedoutonaC18HPLCcolumnusingagradientflowofmobilephase(0.01mol/Lmonobasicpotassiumphosphate(pH3.20)-acetonitrile,63∶37,v/v).Fluoresceinisothiocyanatewasusedasinternalstandard.Thedynamicrangeofassaywas3to50ng/mL.Theintradayprecisionwaslessthan10%andaccuracyrangedfrom95.2%to112.6%.Thesameforinterdaycheckwaslessthan12%and92.8%to105.1%,respectively.Thedrugwasfoundtobestableundertheassayconditions.Thedevelopedmethodissimple,precise,accurate,andstable.Thisindicatesthatitcanbeappliedtoroutineanalysisofthisdruginhumansubjectswheretherearelargenumbersofsampleswithouttheneedofspecializedinstrumentslikecolumnswitching.
